Our organisation does not use cloud infrastructure and we would like to know how we can support our in house server architecture. Please respond with any helpful suggestions which could be used.

Yes, Microsoft Defender Antivirus is already built-in to Windows Server 2016 and higher operating systems. For older operating systems such as Windows Server 2012 R2 and older, you will need to use System Center Endpoint Protection (SCEP) for Antivirus, and then for EDR you can either deploy the MMA agent as described here https://www.thecloudtechnologist.com/defender-for-endpoint-mdatp-for-windows-servers/
